Plasmid encoded lacticin 481 production in Lactococcus lactis subsp. lactis CM41

Abstract

Twenty L. lactis subsp. lactis strains, isolated from raw milk and whey samples, were screened for their antimicrobial activity. One of the strains was selected for its broad activity spectrum of inhibition against various food pathogens, including Listeria and Clostridium strains. Proteolytic enzymes confirmed the proteinaceous nature of the substance. Bacteriocin was purified by ammonium sulphate, ion exchange and reverse phase chromotography. Amino acid sequence showed strong homology with lacticin 481. This result was confirmed by PCR amplification and DNA sequence analysis. Lacticin 481 was encoded by a 52.0 kb conjugative plasmid in L lactis subsp. lactis CM41.
